If you've updated the game recently, I would start with the firewall settings. Make sure turbinelauncher.exe and ddoclient.exe are not blocked.

Do you have windows 10? There's another post about that from Cordovan today or yesterday that gives the link to download additional files to get DDO to work on windows 10.

Try right clicking the launcher and running it as admin. I had to do this once when upgrading to Win10 way back.
