Item Defence On Multiclass Characters

Does Item Defence stack when you have two classes that have it available? That is, for example, if say a Fighter/Rogue took both Item Defence I for 25% chance of less damage to items does that give them 50%?

Usually like bonus's from different classes are locked once you take the 1st level. For example if you take fighter haste boost you are no longer able to take rouge haste boost. I expect its the same for item defense.

This is correct. If the OP was to take the Item Defense I enhancement for one class it would lock out the ID enhancements for all the other classes he/she has on that character. This would include the higher tiers from the class he/she didn't select in the first place.
