Yes. Every time you earn a level you can decide in which class you take that level. So you can create anything from a Rogue19/Barb1 to Rogue1/Barb19. Just talk to the appropriate trainer when you earned a level.
But make sure you know what you're doing. Think about which class and how many levels of it to splash.
Note: DDO limits you to a maximum of three different classes. E.g. you could build a Rogue1/Ranger1/Barb1 but not a Rogue1/Ranger1/Barb1/Fighter1
For each level you gain, you can pick which class to level in, up to a limit of 3. So at level 2 you would be a Rogue 1/ barbarian 1 and at level 3 you could be a Rogue 2/ Barbarian 1, a Barbarian 2/ Rogue 1, or even a Rogue 1/ Barbarian 1/ Ranger 1.
Now you don't want to do this without a plan. Look at what each level of your classes gives you and see what you gain and what you loose. For Rogue/ Barbarians, it's usually 1 or 2 levels of Barbarian for Martial Weapons, hp and a Rage or 2 with the rest Rogue or 2 levels of Rogue for Evasion and a few Rogue skills(OL and UMD) to go with 18 levels of Barbarian. Rogue 13/ Ranger 6/ Barbarian 1 or Barbiarian 12/ Ranger 6/ Rogue 2 also have their fans.
Be very careful when multi-classing.
You get some short term gain but it may bite you in the pants down the track.
I sat down and designed a Bard/Fighter/Rogue (endpoint is 16/2/2). He's at 13/2/2 and his DPS is better than expected. Rogue skills & UMD where I thought they would be BUT his healing is really bad.
Given I originally thought he could be a secondary healer this has been an unpleasant suprise.
I'm going to respec his enhancements totally towards DPS/Buff and just scroll heal off-combat (except for self healing).
If you are going to multi really consider the consequences all the way to L20.
