If you connect using an internet proxy (setup in Internet Explorer) you can check this by clicking on
Tools > Internet Options > Connections (tab) > LAN Settings (button), look to see if "Use a Proxy" is enabled and if it is first check with your ISP to confirm that the settings are a.) correct and b.) necessary for you to connect (some malware/virus' set themselves up as a proxy on the system).
If the proxy setting is correct and needed, renaming the Userpreferences.ini file (eg. olduserpreferences.ini) in your "My Documents" (XP) or "Documents" (Vista) > "Dungeons & Dragons Online" should have the game redetect it and set it up but if not you might need to edit the prefs file manually.
If you do use a proxy server make a note of what it is in I.E. (name and port) and then open the Userpreferences.ini file and add it to the following lines if after renaming it and relaunching the game doesn't do it automatically to the new one it creates on exit:
