For example, do you need to be in a primal sphere destiny to use Perfect Two Handed Fighting? Or can you grab it while being in LD?

You have to have maxed out a Destiny in the Primal sphere, full xp earned all the way to the end, with no more xp possible to be earned in that Destiny.

As long as you meet that criteria it doesn't matter what Destiny you are currently in. You could be in Magister for all the system would care.
