How many points/ranks do I need to get to lv 2. I have passed the first five ranks and I'm still at lv 1.

Where is the Sanctuary where the girl is held and needs to be rescued?

When you have enough experience points to level, you go see your trainer (in aspirant's corner, outside the weapon shop in the harbor) and you train for level 2. Since you can train any class for your character's level 2, they don't make the choices for you, but let you train any class you'd like. If you are playing a Barbarian, and you'd like to get a couple of extra feats, you can take two levels of Fighter and then go back to training Barbarian levels. If you wanted to play a Wizard, but have disabling and open locks skills, you could take the first level as a rogue, and then take 15 levels of Wizard. Many people do, and they succeed as rogues and casters at the same time.

The Haunted Warehouse (for the mission you get in the Leaky Dinghy from the the representative of House Phiarlan) is just outside the apothecary shop in the harbor.

The Sanctuary (for the mission from Friar Renau in the marketplace) is inside the catacombs, behind the Friar, down one flight of stairs and make a left.

If you've passed all five ranks as you state, then your experience bar on the bottom of the screen should be blue and you should have a blue level-up icon in the upper right corner of your screen. At this time, you need to find a trainer of the class you wish to add or advance in, and take your level. It's not automatically granted once you achieve the necessary experience, allowing you to choose which class you take your next level in.

This question is unclear. What is the name of the girl? Where did you pick up the quest? There are many damsels in distress in this game and there are actual locations called Sanctuary, so which is it. I suspect it's either Arissa (Sewer Rescue) or the House J ward (Missing Ward). Both are in Philosopher's Row. Your map should have chalice icons indicating quest entrances once you've picked them up. Hope this helps.
